We wanted to make some adjustments to the column headers on our sheets that are connected via Sheetgo and wanted to make sure it would not interrupt the workflow and connections.

It depends on the adjustments you want to make.
If you are just renaming your column headers, those changes should sync without affecting anything on your destination.
If, however, you want to remove/add columns, you need to first check if anything on the destination depends on those.
In general, additional columns should always be appended at the end to least interfere with the set up workflow.
